The aim of the study was to analyze the nanomechanical properties of C. albicans cells derived from the oral cavity of healthy people in a biofilm produced in laboratory conditions. Candida spp. were sampled from the oral cavity of healthy individuals. The process of biofilm formation was analyzed using classic microscopic observation enriched with SEM (scanning electron microscope) and the nanomechanical properties of the cells were assessed with the use of the atomic force microscopy technique (AFM). From all isolated strains in the samples collected of the oral cavity healthy people was detected 79% C. albicans. Other isolated species belonged to the group "non-albicans". The observations of C. albicans carried out in 24-h cultures revealed a tendency of the cells to form a biofilm structure with multilayer cell systems. The diameter of C. albicans cells in this structure was 5.75 ľm, and the length of the pseudohyphae was 17.08 ľm. The presence of an extracellular substance surrounding the C. albicans cells was detected. The mean value of the adhesion force determined for C. albicans cells was 4.01 nN. Areas with increased hardness (Force Modulation Mode signal; FMM signal) were found mainly in the zones of cells in contact with the glass substrate. The analysis of Candida cells in liquid samples gives satisfactory results, as it prevents unfavorable changes in the cell surface and thus provides more reliable results. The quality of the biofilm is probably related to the nanomechanical properties of C. albicans cells and may consequently contribute to the stability of the biofilm structures and their susceptibility or resistance to antifungal drugs. The presence of Candida spp. especially in companion animals (dogs, cats) poses a risk of their transmission to the human organism. For this reason, it is advisable to undertake additional research to analyze the ability of zoonotic-origin Candida spp. to form biofilms with comparison of the biofilm-formation capacity of species isolated from humans. The exact time point of Treg emergence is still disputed. Furthermore, the time of Treg-mediated suppression mechanisms' emergence has not been identified. It is not yet known whether Treg suppression mechanisms diverged from a single pathway or converged from several sources. We investigated the evolutionary history of Treg suppression pathways using various phylogenetic analysis tools. To ensure the conservation of function for investigated proteins, we augmented our study using nonhomology-based methods to predict protein functions among various investigated species and mined the literature for experimental evidence of functional convergence. Our results indicate that a minority of Treg suppressor mechanisms could be homologs of ancient conserved pathways. For example, CD73, an enzymatic pathway known to play an essential role in invertebrates, is highly conserved between invertebrates and vertebrates, with no evidence of positive selection (w = 0.48, p-value < 0.00001). Our findings indicate that Tregs utilize homologs of proteins that diverged in early vertebrates. However, our findings do not exclude the possibility of a more evolutionary pattern following the duplication degeneration-complementation (DDC) model. Ancestral sequence reconstruction showed that Treg suppression mechanism proteins do not belong to one family; rather, their emergence seems to follow a convergent evolutionary pattern.

It is closely related to the increasing frequency of ticks. The aim of the study was to identify Borrelia burgdorferi, Anaplasma phagocytophilum, Francisella tularensis, and Coxiella burnetii in ticks from selected regions of north-eastern Poland. The material for molecular tests were ticks belonging to two species: Dermacentor reticulatus (meadow tick) and Ixodes ricinus (common tick), collected in the period from July 2020 to June 2021. The studies were performed on arachnids collected mostly from the ground (90.4%), as well as removed from dogs (8.4%) and humans (1.2%). The genetic material was amplified by PCR in the multiplex PCR variant (B. burgdorferi and A. phagocytophilum) and single PCR (F. tularensis, C. burnetii). Only 9 out of 500 ticks tested showed genetic material indicating infection with the above-mentioned microorganisms (1.8%). D. reticulatus ticks dominated in this group. The highest number of ticks, that is seven D. reticulatus and one I. ricinus, were infected with B. burgdorferi. Only one I. ricinus tick (0.3%) and one D. reticulatus tick (0.2%) carried genetic material of A. phagocytophilum. In the latter case, it was a co-infection with B. burgdorferi. Tests on ticks showed a low degree of infection with B. burgdorferi and A. phagocytophilum bacteria (1.8%) and no genetic material of F. tularensis or C. burnetii. On the other hand, one D. reticulatus tick was coinfected with B. burgdorferi and A. phagocytophilum (0.2%). The low percentage of infected I. ricinus ticks may have been due to the small number of specimens examined, which in turn resulted from the fact that D. reticulatus was the dominant tick species in the research area.

In Poland, the most common species of these arachnids are Ixodes ricinus (castor bean tick) and Dermacentor reticulatus (ornate cow tick). There are risks associated with the parasitic effects of ticks. During their feedings, they can have direct and indirect effects on the affected host. Both arthropod species are characterized by a large number of species of microorganisms they transmit causing bacterial, viral, and parasitic diseases. Bacterial infections are the most common among tick-borne diseases. These include Lyme disease, human granulocytic anaplasmosis, tularemia, and Q fever. An increasingly common clinical issue prompting the current research is co-infections, i.e. simultaneous infection of the host by several pathogens. An infection caused by multiple pathogens results in severe and atypical symptoms, prolonged duration of disease, as well as reduced response to treatment. Taking into account the ubiquity of ticks in the environment, only a thorough understanding of tick-borne diseases, including infectious agents and vectors, can lead to better control of this problem worldwide. Streszczenie: Kleszcze oraz przenoszone przez nie patogeny wciąż stanowią aktualny problem zdrowotny o charakterze globalnym. W Polsce najbardziej rozpowszechnionymi gatunkami tych pajęczaków są kleszcze Ixodes ricinus (kleszcz pospolity) i Dermacentor reticulatus (kleszcz łąkowy). Możliwe zagrożenia związane są z pasożytniczym oddziaływaniem kleszczy. Ich żerowanie wywiera bezpośredni i pośredni wpływ na zaatakowany organizm. Oba gatunki stawonogów charakteryzują się dużą liczbą gatunków przenoszonych mikroorganizmów, wywołujących choroby bakteryjne, wirusowe oraz pasożytnicze. Infekcje bakteryjne są najbardziej liczną grupą spośród chorób odkleszczowych, Zalicza się do nich m.in. boreliozę, anaplazmozę granulocytarną, tuleremię i gorączkę Q. Coraz częściej pojawiającym się zagadnieniem klinicznym, stanowiącym aktualny problem badawczy, są koinfekcje, czyli współzakażenie organizmu kilkoma patogenami. Zakażenie spowodowane przez różne patogeny skutkuje występowaniem nasilonych i nietypowych objawów, wydłużonym czasem trwania choroby, jak również brakiem odpowiedzi organizmu na zastosowane leczenie. Biorąc pod uwagę wszechobecność kleszczy w środowisku, tylko dokładne zrozumienie chorób odkleszczowych, w tym czynników zakaźnych oraz wektorów, może doprowadzić do lepszej kontroli tego problemu na całym świecie.

The spread of D. reticulatus enhances the risk of exposure of domestic animals and their owners to tick-borne diseases. The objective of this study was to assess the prevalence of infection of D. reticulatus ticks feeding on dogs with the pathogens Borrelia burgdorferi sensu lato and Anaplasma phagocytophilum. The study material comprised 152 D. reticulatus ticks collected from dogs in the northeastern part of Lublin Province (eastern Poland). A ready-made AmpliSensŽ TBEV, B.burgdorferi sl, A.phagocytophilum, E.chaffeensis/E.muris-FRT PCR kit was used for qualitative detection and differentiation of tick-borne infections. The assessment of the degree of infection of the analyzed ticks with the two pathogens revealed that 9.2% (14/152) of the examined ticks were infected with one of the pathogens. No co-infections with the pathogens were detected in any of the ticks. The highest specific percentage of infections (8.6%, 13/152) was associated with A. phagocytophilum. The presence of B. burgdorferi s.l. was detected in only one of the examined ticks (0.7%). The spread of D. reticulatus to new territories and the increase in population density in areas of their regular occurrence implies the need for further studies of the prevalence of pathogens with medical and veterinary importance in order to assess the risk of tick-borne diseases. The proportion of borreliosis infections caused by B. burgdorferi spirochaetes is increasing. Difficulties diagnosing this disease with conventional methods have led to growing molecular biology research aimed at developing improved diagnostic tools. Molecular biology methods include all techniques that allow the detection and analysis of nucleic acids. Among them there are methods based on PCR reaction, molecular typing methods (MLST), new generation sequencing (NGS). The current development of this field gives great research opportunities. With regard to B. burgdorferi spirochete, molecular biology is used to: confirm Lyme borreliosis, identify and type Borreliella spirochetes, detect them in tick vectors or intermediate hosts, as well as to identify co-infections between different Borreliella species and other tick-borne pathogens. They are meant to significantly improve diagnostic analyzes. This paper reviews the current methods used for the detection and identification of B. burgdorferi. Molecular mechanisms for the survival of spirochetes in the host, infection vectors and clinical picture of Lyme disease were also discussed. Streszczenie: Borelioza, jako jedna z chorób odkleszczowych, od kilku lat stanowi aktualny problem epidemiologiczny w Polsce i na świecie. Wzrastający odsetek przypadków zakażeń boreliozą, chorobą wywołaną przez krętki B. burgdorferi, bardzo trudnych do zdiagnozowania metodami konwencjonalnymi, powoduje stale powiększające się zainteresowanie najnowszymi technikami zdziedziny biologii molekularnej. Metody biologii molekularnej obejmują wszystkie techniki pozwalające na wykrywanie i analizę kwasów nukleinowych. Wśród nich wyróżnia się metody oparte o rekację PCR, metody typowania molekularnego (MLST), sekwencjonowanie nowej generacji (NGS). Aktualny rozwój tej dziedziny daje ogromne możliwości badawcze. W odniesieniu do krętków B. burgdorferi biologię molekularną wykorzystuje się do: potwierdzenia boreliozy z Lyme, identyfikacji i typowania krętków Borreliella, wykrycia ich w wektorach kleszczowych, czy gospodarzach pośrednich, a także przy stwierdzeniu koinfekcji między różnymi gatunkami Borreliella i innymi patogenami przenoszonymi przez kleszcze. Mają one w znacznym stopniu usprawnić analizy diagnostyczne. W niniejszej pracy przedstawiono przegląd wszystkich metod umożliwiających wykrycie i identyfikację B. burgdorferi. Omówiono również molekularne mechanizmy umożliwiające przetrwanie krętków w organizmie gospodarza, wektory zakażenia oraz obraz kliniczny boreliozy.

The aim of this study is a general review on the possibilities of counteracting Lyme disease, and efforts to combat the disease in the light of recent scientific findings. Fundamental survival strategies of living organisms include mutual interaction, independent of the elements that form it, cell structure complexity, size, or mass, as well as the ability to select the right countermeasures to vector-borne diseases. This paper specifically presents the issues of dependence and interrelationships between humans, ticks, and Lyme spirochete through a detailed presentation of the problem of Lyme disease in athletes. Furthermore, this article presents strategies used by the Lyme spirochete to infect the host's organism, strategies used by the arachnid to cause infection and its preferences in choosing a host, and principles of modern microbiological diagnostics and prophylaxis as a tool in the fight against a tick infected with B. burgdorferi.
